High-throughput organelle patch clamp: overcoming isolation and scalability bottlenecks
Speaker: Bastien Masson, ORIA Bioscience
Patch-clamp electrophysiology on organelles remains technically demanding: failed preparations, high variability, and incompatible toolchains slow down even experienced teams. Organelles concentrate this challenge, sitting at the intersection of high pharmacological relevance and notoriously difficult access. In this webinar, ORIA Bioscience and Nanion Technologies show how combining ready-to-use, purified lysosomes and mitochondria with automated patch-clamp platforms can fundamentally change the game. We'll cover how to streamline your preparation workflow, reduce assay variability, and generate high-quality organelle electrophysiology data more consistently.
Approaching organelles with high-throughput electrophysiology Methods to allow early Drug Discovery from native Lysosomes
Speaker: Markus Rapedius, Nanion Technologies
Lysosomal ion channels and transporters have emerged as pivotal components in the regulation of cellular homeostasis and intracellular signaling pathways, linking lysosomal function to diseases such as Parkinson's and cancer. Consequently, their potential as therapeutic targets have driven the demand for high-throughput electrophysiological methods. However, despite their recognized importance, direct functional analysis of these channels within native lysosomes has remained a significant technical challenge, as traditional manual patch-clamp methods are time-consuming and low in throughput. While automated patch clamp systems have been active for many years for whole-cell patch clamp, we have now tailored our automated high-throughput patch clamp platform (Nanion's SyncroPatch384) for the study of ion channel activity in native lysosomal preparations obtained from ORIA Bioscience as well as from a modified differential ultracentrifugation approach. This methodology enables rapid, reproducible, and scalable measurement of lysosomal ion channels, thereby facilitating comprehensive investigations into lysosomal function in both health and disease contexts, and advancing applications in drug discovery.
