They are members of\u00a0the sodium-selective cation channels belonging to the\u00a0epithelial sodium channel\/degenerin (ENaC\/DEG)\u00a0family. ASICs are highly sensitive to extracellular acidosis\u00a0and are permeable to cations, predominantly Na<sup>+<\/sup>.\u00a0So far, 6 different ASIC subunits have been identified\u00a0encoded by 4 genes. They are found expressed throughout\u00a0the CNS and PNS and have a proposed role in\u00a0nociception and pain, and other neurological\u00a0diseases such as ischaemia and inflammation. The ASIC3\u00a0channel was first identified in the late 1990\u2019s. It was\u00a0found to be localized to primary afferent nociceptive\u00a0fibers innervating the skin, muscles, joints and viscera, in\u00a0agreement with a role in pain perception. Furthermore,\u00a0ASIC3 is expressed in higher amounts in nociceptive\u00a0neurons innervating muscle (~ 50%) compared to skin\u00a0(~ 10%), which indicates that ASIC3 may play an\u00a0important role in detecting muscle acidosis.\u00a0Here we present high quality data at a high throughput\u00a0collected on the SyncroPatch 384PE showing\u00a0activation and inhibition of ASIC3 expressed\u00a0in HEK cells. The pH which elicited a halfmaximal\u00a0response was in good agreement with the\u00a0literature. The IC<sub>50<\/sub>\u00a0for block of the ASIC3 current\u00a0by amiloride, a known blocker of ASIC and ENaC\u00a0channels, was also in good agreement with the\u00a0literature.
