{"id":13287,"date":"2023-05-18T17:15:51","date_gmt":"2023-05-18T17:15:51","guid":{"rendered":"https:\/\/www.nanion.de\/?post_type=resource_library&#038;p=13287"},"modified":"2024-01-09T17:17:07","modified_gmt":"2024-01-09T17:17:07","slug":"2023-predicting-herg-repolarization-power-at-37c-from-recordings-at-room-temperature","status":"publish","type":"resource_library","link":"https:\/\/nanion.com\/ja\/resource_library\/2023-predicting-herg-repolarization-power-at-37c-from-recordings-at-room-temperature\/","title":{"rendered":"2023 &#8211; Predicting hERG repolarization power at 37\u00b0C from recordings at room temperature"},"content":{"rendered":"<p>Loss-of-function and gain-of-function mutations in the\u00a0<i>KCNH2<\/i> gene cause long and short-QT syndromes (LQTS or SQTS), respectively, predisposing to life-threatening cardiac arrhythmias.\u00a0<i>KCNH2<\/i>\u00a0encodes the voltage-gated K<sup>+<\/sup>\u00a0channel hERG that generates the delayed rectifier K<sup>+<\/sup>\u00a0current I<sub>Kr<\/sub> controlling the action potential (AP) duration.\u00a0Prolonged or shortened ventricular AP durations are visualized as abnormal QT interval duration on the electrocardiogram. The occurrence and severity of\u00a0<i>KCNH2<\/i>-related arrhythmias are determined by the variant functional impact. Sequencing\u00a0<i>KCNH2<\/i>\u00a0has provided a plethora of variants associated or not with pathological cardiac phenotypes and indexed in the ClinVar NCBI database. Discriminating pathogenic variants from benign ones would clarify the genetic background of patients and relatives, and stratify the risk of adverse events. In the face of a wide spectrum of hERG functional defects, we looked for a way to summarize the net loss or gain of function in a unique index.
